چکیده

Ca(2+)-dependent exocytosis of synaptic vesicles is mediated by the SNARE proteins synaptobrevin/VAMP, SNAP-25, and syntaxin. SNARE function is controlled by conserved regulatory proteins, including the complexins. In a study by Xue et al. in this issue of Neuron, contradictory data from Drosophila and mouse complexin mutants have been resolved, revealing a complex pattern of facilitatory and inhibitory domains.

Complexin: Does It Deserve Its Name?

Knockout and other perturbations of complexins have provided important insights and elicited controversies about their role in neurotransmitter release. New work by Yang et al. in this issue of Neuron adds important detail and complexity to existing concepts-particularly on the nature of a Ca(2+)-dependent complexin-synaptotagmin switch for the triggering of exocytosis. But it also provokes tho...

Tilting the Balance between Facilitatory and Inhibitory Functions of Mammalian and Drosophila Complexins Orchestrates Synaptic Vesicle Exocytosis

SNARE-mediated synaptic exocytosis is orchestrated by facilitatory and inhibitory mechanisms. Genetic ablations of Complexins, a family of SNARE-complex-binding proteins, in mice and Drosophila cause apparently opposite effects on neurotransmitter release, leading to contradictory hypotheses of Complexin function. Reconstitution experiments with different fusion assays and Complexins also yield...

Complexin activates exocytosis of distinct secretory vesicles controlled by different synaptotagmins.

Complexins are SNARE-complex binding proteins essential for the Ca(2+)-triggered exocytosis mediated by synaptotagmin-1, -2, -7, or -9, but the possible role of complexins in other types of exocytosis controlled by other synaptotagmin isoforms remains unclear. Here we show that, in mouse olfactory bulb neurons, synaptotagmin-1 localizes to synaptic vesicles and to large dense-core secretory ves...

A Complexin/Synaptotagmin 1 Switch Controls Fast Synaptic Vesicle Exocytosis

Ca(2+) binding to synaptotagmin 1 triggers fast exocytosis of synaptic vesicles that have been primed for release by SNARE-complex assembly. Besides synaptotagmin 1, fast Ca(2+)-triggered exocytosis requires complexins. Synaptotagmin 1 and complexins both bind to assembled SNARE complexes, but it is unclear how their functions are coupled. Here we propose that complexin binding activates SNARE ...

The Place-Name as an Intangible Place of Memory (A Holistic Approach in Reading the Place-Names through a Comparative-Analytical Study on the Character of Name and Place)

Understanding architectural heritage and their various aspects have always been a subject of focus for the international conservation communities. Within the recent decades, eventhough the place-names are part of the living history as well as cultural heritage, they have still constantly been facing quick precipitant changes. As such, in the Conservation literature, most studies have skipped ad...
